I think this might be related to the issue of flash support or embedded objects. The following script should be added to a link to make it to where you can add a call me button to your signature.
<embed src="http://embed.grandcentral.com/webcall/[REMOVED FOR PRIVACY]" width="142" height="54" wmode="transparent"></embed>
The signature preview show this properly but the actual email signature doesn't work.

Once again, this is almost certainly related to the way Gmail handles code. I am virtually certain that it strips any <embed> or <object> tags, as it would do as well with any <script> tags. This is due to the security and process that Gmail itself has implemented and is something over which the add-on has no controll.

Great add-on.
One bug that I have noticed is that I can't play YouTube videos (or any other flash videos) with GMail Signatures installed. This also applies to playing my Grand Central messages. When I disable GMail Signatures and restart Firefox, no problem.

Odd, I'm sorry you're having this kind of conflict. I've never run into this kind of problem myself, so it may be a little hard to troubleshoot.
